Immunocytochemistry:
1. Fix cells with a 4% Paraformaldehyde solution for 30-45 minutes, and then rinse several times with Tris-Buffered-Saline (TBS).
2. Incubate cells with MAB4301X diluted to 1/100 with PBS/ Tween-20 (0.05%) / BSA (1%) for 30 minutes.
3. Wash the sample three times with PBS / Tween-20 (0.05%) for 1 minute per wash. Examine under a microscope fitted with a FITC filter.
Optimal working dilutions must be determined by end user.
This Anti-Stage-Specific Embryonic Antigen-1 Antibody, clone MC-480, Alexa Fluor 488 is validated for use in FC, IC, IF for the detection of Stage-Specific Embryonic Antigen-1.

General description
SSEA-1 is expressed on the surface of early mouse embryos, murine embryonal carcinoma cells (EC), murine embryonic stem cells (ES) and murine & human germ cells (EG). No immunoreactivity is evident with undifferentiated human EC and ES cells. Expression of SSEA-1 is down regulated following differentiation of murine EC and ES cells. In contrast, the differentiation of human EC and ES cells is characterized by an increase in SSEA-1 expression.
Immunogen
F9 teratocarcinoma stem cells (X-irradiated)

Purified immunoglobulin conjugated to Alexa Fluor® 488. Liquid in Phosphate buffer with 15 mg/mL BSA as a stabilizer and 0.1% sodium azide
Specificity
Reacts with the Stage-specific embryonic antigen-1 (SSEA-1) that is expressed upon the surface of early mouse embryos, murine embryonal carcinoma cells (EC), murine embryonic stem cells (ES) and murine & human germ cells (EG). No immunoreactivity is evident with undifferentiated human EC and ES cells. Expression of SSEA-1 is down regulated following differentiation of murine EC and ES cells. In contrast, the differentiation of human EC and ES cells is characterized by an increase in SSEA-1 expression.
